Adapting fish defenses to block human infections: Antimicrobial peptide of fish gills inspire clean surfaces

ScienceDaily (Nov. 29, 2012) ? An undergraduate research team analyzes peptides from fish gills to engineer antimicrobial surfaces for food preparation and medical devices.

Living in an environment teaming with bacteria and fungi, fish have evolved powerful defenses against waterborne pathogens, including antimicrobial peptides located in their gills. Undergraduate researchers at Worcester Polytechnic Institute (WPI) are studying the biology and the mechanics of one of those peptides with the hope they can use that knowledge to create engineered surfaces that kill bacteria responsible for foodborne illnesses and hospital-acquired infections.

The research team, led by Terri Camesano, professor of chemical engineering, reports its latest findings in the paper "Creating Antibacterial Surfaces with the Peptide Chrysophsin-1," published online in October by the journal ACS Applied Materials & Interfaces.

"Fish have a wonderful solution for blocking bacterial and fungal infections," Camesano said. "In this study, we are working to better understand the biochemical mechanics of that process."

As fish filter water through their gills to extract oxygen, antimicrobial peptides (AMPs), including Chrysosphin-1, trap and kill pathogens before they can invade the fish's bloodstream. Scientists in many laboratories around the world are actively exploring the potential use of these molecules to prevent human infections. In the current study, the WPI team attached AMPs to silicon and gold surfaces using two different approaches and measured the bound peptides' ability to kill the bacterial pathogen E. coli.

In the first method, the AMPs were absorbed directly onto gold and silicon crystals, forming a single layer of molecules with the AMPs lying flat on the surface. In the second method, the tips of the AMPs were attached to the surfaces with a glue-like substance so that the peptides rose vertically, like blades of grass extending up from the ground. Surfaces with both AMP configurations were cultured with E. coli cells. The results showed that when the AMPs were lying flat they killed 34 percent of the bacteria in the culture, but when they were standing up vertically they killed 82 percent.

"The hypothesis is that when peptides are attached vertically to the surfaces, they are better able to move and bend so they take on a shape that is more effective in binding to and disrupting the E.coli cells," Camesano said.

In addition to gathering data about the antibacterial efficacy of the attached AMPs, the WPI research team developed a technique for monitoring, in real time, the attachment of AMPs to surfaces. Using quartz crystal microbalance with dissipation monitoring (QCM-D), the team measured the quantity of AMPs that successfully attached to the surfaces in the horizontal and vertical orientations and the density of the AMP layers, along with other properties.

"This was a powerful process, to be able to essentially watch the binding process as it happened," Camesano said. "It is a technique that we will continue to apply in further studies."

Camesano said gold and silicon surfaces were selected for the current study because their chemical properties are well-suited for AMP binding. In ongoing work, Camesano's laboratory will continue to characterize the mechanics of AMP binding for optimal antimicrobial activity and test other materials, including titanium, stainless steel, and plastics, that would have greater utility in food preparation and healthcare.

"What is also notable about this study is that it is the work of undergraduates," Camesano said. "They've done excellent work here that will inform future graduate studies in our lab."

The WPI undergraduates who co-authored the AMP paper are Ivan Ivanov '12 (chemical engineering), Alec Morrison '12 (biochemistry), and Jesse Cobb '12 (chemical engineering). Co-author Catherine Fahey, a student at George Washington University, worked on the study during at a summer project at WPI. Their work was sponsored in large part by a grant from the National Science Foundation's Research Experiences for Undergraduates (REU) program.

Mississippi's only abortion clinic fights to stay open

Mississippi's only abortion clinic will have to close unless a federal judge halts a new state law requiring its physicians to obtain admitting privileges to local hospitals, according to a court motion filed on Wednesday.

In the motion, the Jackson Women?s Health Organization renewed its request for a federal judge to prevent state officials from enforcing a law which went into effect on July 1.

Under the new law, the clinic must hire a doctor who is a board-certified OB-GYN and who also has admitting privileges to a local hospital where patients may be referred if further treatment is needed.

The clinic's providers are board-certified OB-GYNs, but the only one with admitting privileges when the law passed provides limited service at the clinic. Currently on the clinic?s website is a statement seeking a doctor who could perform abortion care for patients up to 16 weeks in the pregnancy.

Mississippi, which had as many as 14 abortion providers in the early 1980s, has some of the country's strictest abortion laws and one of the lowest abortion rates. It also has the highest teen pregnancy rate in the United States - more than 60 percent above the national average in 2010.

In 2010, the latest year for which figures are available, 2,297 abortions were performed in Mississippi with no deaths and one complication, according to the health department.

Any facility in Mississippi that performs 10 or more abortions a month or 100 or more in a year needs a license from the state. The Jackson Women?s Health Organization in Jackson has been the sole licensed abortion clinic in the state since 2002.

Supporters of the law have made it no secret they would like to end all abortions in Mississippi, although they have also said it was designed to protect women?s health.

"It is now clear that plaintiffs have no hope of being able to comply with the Admitting Privileges Requirement," said the filing by the New York-based Center for Reproductive Rights, which represents the clinic in court.

The state Department of Health, tasked with enforcing the law, gave the clinic until January to comply.

But clinic owner Diane Derzis told Reuters she sent applications on behalf of all the physicians to every hospital within a 30-mile radius. All of the hospitals ultimately rejected the requests or refused to even consider them, she said.

Jackson Women's Health Organization, based in the city of Jackson, has operated in the state for 17 years and has been Mississippi's only clinic since 2002. Its closure would mean an end to elective abortion in the state and would force women to travel elsewhere for the procedure.?

Police told NBC News that they?arrested Matthew Allen Cook on Oct. 31, after the stolen laptop showed up at a pawn shop and employees identified the suspect. But police said?that?they are?leaving the issue of how he entered the rooms to the prosecution.

The strange circumstance of a locked?door being opened without a key would be a mystery worthy of Sherlock Holmes, if not for one thing: The lock was one with a well-known security exploit made public by a hacker earlier this year.

The lock in question is from Onity, a major supplier of electronic and keycard locks for hotels like the Hyatt. Cody Brocious, a software engineer at Mozilla and hobbyist hacker, demonstrated a vulnerability in many of their locks in July, afterwards showing a refined technique onstage at the Black Hat hacker conference. Andy Greenberg at Forbes reports that the method used in the recent break-ins is more than suggestive of the hack.

Keycard locks like those in hotels often have a port on the bottom used to access the lock's electronic memory or activate it when its battery has run out. Brocious showed that the security inside the lock is extremely easy to circumvent using a cheap and portable device made with off-the-shelf parts. It was unreliable at first, but other hackers have improved the technique and even fit the device into the shell of a dry-erase marker.

Onity, for its part, has acknowledged the flaw but offers only a temporary solution: blocking the port with a bit of plastic or putty. The flaw is baked into the device's firmware, and the only way to fix it permanently is to replace it entirely, which could be costly with at least?hundreds of thousands of locks affected by the flaw. The Hyatt hotel in question has opted to glue the port shut.

In the meantime, the case against the suspect is progressing in Harris County. But even if he is charged and found guilty, the hack that opens hotel doors around the world is still at large. As Todd Seiders, a security expert at Petra Risk Solutions, told Forbes: "We?re expecting incidents in which these devices are used to explode nationally...?We?re going to get hit hard over the next year."

S.African stocks end lower on US fiscal worries

JOHANNESBURG (Reuters) - South African stocks ended down on Wednesday after worries about the U.S. economy and concerns over Greece's new debt deal hit the resource-heavy bourse and dragged gold companies such as Harmony Gold sharply lower.

The Top-40 index closed down 1.06 percent at 33,225.91, while the broader All-share index shed 0.97 percent to 37,438.41.

Harmony, South Africa's third-largest gold producer, led the losers, falling 3.85 percent to 70.94 rand after the spot bullion price shed more than 1.5 percent.

Larger peers AngloGold Ashanti and Gold Fields also closed in negative territory, down 2.4 percent and 2.5 percent, respectively.

Investment holding company Remgro ended down 0.9 percent after warning it was likely to report a more than 20 percent drop in first-half earnings.

Gainers were led by base metals miner Assore, up 1.9 percent on the day, followed by consumer goods group Tiger Brands, up 1.62 percent.

Shares of Amalgamated Appliance Holdings closed up 6.6 percent after conglomerate Bidvest said it would buy the remaining 72 percent stake in the distributor and retailer of household appliances it does not already own in a $61 million cash deal.

Van Eck said he expected the negative sentiment to hold through the rest of the week.

"I'm still waiting to see this market come off a bit easier," he said. "There is still a bit of a bubble that is sitting there in the retail sector."

Retailers, which include Massmart, Shoprite, Pick n Pay and Spar, are among the best performing stocks in Johannesburg this year.

Trade volume was reasonable, with just under 165 million shares changing hands, compared with last year's daily average of 255 million shares. Decliners outnumbered advancers at 170 to 116, with 57 stocks unchanged.

Gibbs Sports Amphibians brings PWC/ATV hybrid Quadski to powersports market

The initial feeling is strange. Seconds after driving an ATV into a lake, the vehicle floats. With the push of a button, the wheels retract, and the once looming ATV is transformed into a commanding and stable PWC.

The Quadski, Gibbs Sports Amphibians? first commercially available vehicle, was introduced to select media in mid-October. It was expected to hit powersports dealers? showrooms in mid-November.

The vehicle is an 18-year labor of love for company founder Alan Gibbs, an accomplished entrepreneur hailing from New Zealand. Full development of the amphibian technology began about 15 years ago, when chairman Neil Jenkins, an experienced engineer in the automotive and aerospace industries, came on board and merged his Krafthaus with Gibbs Technologies. More than $200 million and 2 million man hours have gone into the development of Gibbs Sports? High Speed Amphibian (HSA) technology, but the Quadski is finally available at select dealerships, and other amphibian models will join the company?s lineup over the next several years.

A solution to a nagging problem
Gibbs began developing his amphibian technology to solve a personal problem. The company founder owns a farm on Kaipara Harbour in New Zealand, which has up to a mile-long tide. With so much beach between the farm and the ocean, Gibbs had no way to get a boat out to sea.

?If you take a boat out in a tractor and trailer, drop it off, take the trailer, drop in back in the shed, walk back to the boat, it?s 200 yards further out or 200 yards further in,? Gibbs told Powersports Business.

At one point, Gibbs built a catamaran with wheels, but it was too bulky for its intended uses, so he scrapped it and began his journey to develop a practical amphibian vehicle.

In an effort to launch a company to develop amphibian technology, Gibbs flew to Detroit because he thought the U.S. would have a market for amphibians. And, and as Detroit is known as the automotive capital of the country, he thought he could find the necessary engineering workforce there.

?There?s a huge talent pool of people here,? Jenkins explained. ?Almost every supplier on the face of the planet has an office or a representative here, so it made a lot of sense for us to be in this area, try to access those people and the suppliers to make life easier or to do things more efficiently.?

Though the HSA technology has been developed in facilities in Michigan, New Zealand and England, the Quadski is being produced out of the company?s two plants in Auburn Hills, Mich. Gibbs Sports currently has 100 employees in Michigan, and the company plans to add more than 200 jobs in the next year.

In addition to the Quadski, Gibbs Sports Amphibians has about seven vehicles in prototype ? including a side-by-side and a vehicle smaller than the Quadski ? that are expected to hit the market in the next five years. The Phibian, a 30-foot, 4,500-pound, 15-seat amphibian, and the Humdinga, a 21-foot, 6,600-pound vehicle seating five to seven, were introduced to the U.S. government earlier this year. They?ll be distributed through different channels than the powersports vehicles.

The Quadski
Gibbs Sports decided the Quadski would be the first HSA vehicle commercially available because it had the fewest regulatory hurdles to overcome.

?The ATV regulations are much less complicated than the automotive regulations for cars, and as far as the EPA is concerned, it?s treated as a personal watercraft,? Gibbs explained.

The vehicle runs on BMW?s K 1300 175 hp, four-cylinder, water-cooled engine, featuring electronic fuel injection, a double-overhead camshaft and dry-sump lubrication. It has a six-speed gearbox with an automatic transmission that can be shifted at the push of a button.

?You don?t have to use a clutch, and you change gears with a button; it?s all electronic, and if you forget to change gears, it changes them for you,? Gibbs said. ?It?s very sophisticated; our guys did the development of the clutch. The gear change is like when you get in a pedal-shift car, where you push the pedals and go up and down in gears. It?s all automatic.?

The Gibbs Sports Amphibians? Quadski transitions from an ATV to a PWC and vice versa in a matter of seconds.

What makes the vehicle transition easily from ATV to PWC is the always-running jet. The differential is only in operation when land mode is activated. To transition from ATV to PWC, the rider simply enters the water through a boat lift, beach, or other accessible shoreline, drives far enough in to allow the Quadski to float and activates marine mode with a button. Upon exiting the water, the rider presses the button, putting it in land mode and accelerates up the shore.

?When you?re coming out, you have the thrust of the jet as well as the drive of the wheels, so getting out is ? you won?t have a problem getting out. You have the jet thrust and the wheels, so it will switch right up the bank, and the power of the jet combined with the wheels is enough to get you out in just about anything,? Gibbs explained.

Besides conquering the marine and land engine challenge, Gibbs Sports also had to develop a body that would encompass the motor and jet, float on water and handle the rigors of ATVing. The durable, lightweight hull is manufactured from a single-piece mold using current composite technology.

?Everything that we?ve designed on the vehicle we analyzed in terms of making the vehicle as light as we can,? Jenkins explained. ?The body is one part of it; every suspension component, everything has been through that process, so it?s more akin to designing a helicopter in terms of its engineering than it is an ATV, so it?s significant as far as engineering goes.?

Upon seeing the Quadski in person, the first thing anyone familiar with ATVs notices is its large stature. The vehicle is 10.5 feet long, 4.3 feet tall and 5.2 feet wide, with a wheelbase of 5.8 feet. With the size, Gibbs admits it?s not designed to ride on standard ATV trails, but rather, on wider trails or in open areas.

?It?s got different road capabilities from a straight ATV. It?s not a 4-wheel drive; it?s got a much wider track than a conventional ATV,? he said. ?It runs pretty good; it has a pretty wide track, really refined suspension, and it?s terrific going along tracks, rolling across pasture, and you?re going fast.?

As a PWC, the vehicle is broad and stable enough for someone to jump off it into the water. However, it?s not going to corner like a sportier PWC. But Gibbs says the wider body was designed purposefully, to give riders a more stable ride in both modes.

?In just handling a ride, narrow is bad,? he said. ?It can get you down tight trails, but it isn?t good for a vehicle because the wider it is, the more stable, so we?ve never tried to make it narrow because we want it to be a comfortable, quality vehicle.?

The Quadski has been developed to be a good quality ATV and PWC, while not topping the charts in either category.

?I have no aspirations to replace ATVs or PWCs,? Gibbs explained. ?There will be some of that, but basically it will be the customer who can get benefit from our strengths.?

The strengths of the vehicle are its HSA technology, and it?s capability to go 45 mph on land and water.

Dealer network
To distribute the Quadski, Gibbs Sports is developing a dealer network, comprised mostly of powersports dealers. The initial dealers are located in four focus areas ? the Southeast, Northeast, the Great Lakes region and Texas ? but the OEM is willing to talk to anyone who is interested in the vehicles.

?We want dealers to be quality dealers, to have the right quality assessments from other people and to have good relationships with customers, good territories. So we?re not looking to just proliferate dealerships,? Jenkins said.

Gibbs Sports reported that it already has an adequate handful of dealers for the initial launch of the Quadski, but it expects to have a network of 20-30 within the next 12 months. The company plans to produce and sell 1,000 units in the first year.

Each dealer will be required to carry two units ? one for a Gibbs display and another for sale. Only one one-rider version is available for now, and it comes in red, yellow, blue, silver and black.

Gibbs Sports quoted the price at ?about $40,000.? Though that?s significantly higher than any ATV or PWC currently costs, the company needs to recoup some of the R&D and labor costs that have gone into the technology so far. Because of the uniqueness of the model, it?s likely to appeal to those with the means to have one of the first 1,000 production models of this new vehicle.

?It?s like any new technology ? it costs a great deal to get it to this point, but over time the price will come down, and also there will be other models that are inherently cheaper. So initially it?s a high Rolls Royce-type of product, but over time, it will be in the sort of normalized price,? Gibbs explained.

After more than 15 years working on the vehicle, he?s just glad that he can finally bring it to market and begin following up with other HSA models.

?We see it primarily as a fun product,? Gibbs explained. ?You saw the amphibian series work products, but this is fun, and we call it fun squared because it doesn?t just double your fun, it squares your fun, so you go out for a day, and you don?t have to make your mind up whether you?re going to go through trails or if you?re going to go to the water, whatever. You can go out there and have heaps more fun, so this is a fun product. Some people may want to put equipment and stuff on it to make it more useful, like the rescue units and so on, but fundamentally I don?t think I can think of any motorized machine that could give me more fun.?

During his November 14 press conference, President Obama said his broad vision of comprehensive immigration reform ?is very similar to the outlines of previous efforts at comprehensive immigration reform.? This is newsworthy because previous comprehensive immigration reform efforts since 1986 have produced seven amnesties, none of which reduced illegal immigration. This begs the question: Is the permanent reduction of illegal immigration a goal of this administration?

The last bipartisan U.S. Commission on Immigration Reform studied the issue for six years. President Clinton appointed former congresswoman and Democratic icon Barbara Jordan as its chair. Jordan rose from humble means to become a lawyer and the first Southern black woman elected to the House of Representatives. She was a leader in the civil rights movement, a professor of ethics, a recipient of the Presidential Medal of Freedom, and a world-class orator (two of her speeches are considered among the greatest of the 20th century). Her appointment gave the commission instant credibility. Jordan said that she believed her responsibility as the head of the commission was to restore credibility to the U.S. immigration system. On the issue of illegal immigration, Jordan was unambiguous about her goals: ?Unlawful immigration is unacceptable. Those who should not be here will be required to leave.?

President Obama signaled a different philosophy on November 14, saying, ?I do think that there should be a pathway for legal status for those who are living in this country, are not engaged in criminal activity, are here simply to work.?

This is a clear departure from the approach advocated by Jordan, who couldn?t have been clearer on this point. ?There are people who argue that some illegal aliens contribute to our community because they may work, pay taxes, send their children to our schools, and in all respects except one, obey the law,? she said. ?Let me be clear: that is not enough.?

Jordan?s stated concerns were about ?the impact of immigration on the most disadvantaged within our already resident society ? inner city youth, racial and ethnic minorities, and recent immigrants who have not yet adjusted to life in the U.S.? At the time, 3-4 million foreign citizens were working illegally in the U.S. Today, 7 million illegal workers hold U.S. jobs while more than 20 million U.S. citizens and legal immigrant workers can?t find full-time work. If that isn?t the immigration problem that Obama proposes to fix, what is?

Additionally, if Obama believes an exception should be made for illegal aliens who ?are simply here to work,? does that apply only to those who are here illegally today, or will his exception extend to future waves of illegal workers? If he wants to make this exception for any future alien who violates the terms of her visa or crosses the border illegally, why have limits on immigration at all beyond terrorists, rapists, and murderers? If President Obama wants to limit the exception to an eighth (and final?) amnesty, why is he advocating a ?continuation? of policies that got us into this mess in the first place? Obama has rejected Jordan?s framework without articulating an alternative.

Rice meeting with senators fails to dampen criticism

WASHINGTON (Reuters) - U.S. Ambassador to the United Nations Susan Rice on Tuesday failed to win over her harshest Republican critics in the U.S. Senate who are threatening to block her nomination if President Barack Obama chooses her for Secretary of State or another top post in his second-term Cabinet.

Rice met for about an hour behind closed doors at the U.S. Capitol with Senators John McCain, Lindsey Graham and Kelly Ayotte. They have openly criticized her for initial comments after the September 11 attack on the U.S. mission in Benghazi that suggested it was a spontaneous event arising from protests of an anti-Islam film rather than a premeditated attack.

The U.S. ambassador to Libya and three other Americans were killed in the attack on the Benghazi mission and a nearby CIA annex. Intelligence officials later said the attack was possibly tied to al Qaeda affiliates.

"We are significantly troubled by many of the answers that we got, and some that we didn't get, concerning evidence that was overwhelming leading up to the attack on our consulate," McCain told reporters after the meeting.

"It is clear that the information that she gave the American people was incorrect when she said that it was a spontaneous demonstration triggered by a hateful video," he said.

"It was not, and there was compelling evidence at the time that that was certainly not the case, including statements by Libyans as well as other Americans who are fully aware that people don't bring mortars and rocket-propelled grenades to spontaneous demonstrations," McCain said.

Republicans have argued that the Obama administration tried to play down the terrorist angle in its initial comments to avoid undermining the president's claims of success in fighting al Qaeda in the run-up to the November 6 election.

Rice was accompanied by acting CIA Director Michael Morell and was not seen by reporters, but later issued a statement saying: "We explained that the talking points provided by the intelligence community, and the initial assessment upon which they were based, were incorrect in a key respect: there was no protest or demonstration in Benghazi."

"While, we certainly wish that we had had perfect information just days after the terrorist attack, as is often the case, the intelligence assessment has evolved," she said. "We stressed that neither I nor anyone else in the Administration intended to mislead the American people at any stage in this process."

Obama has defended Rice and said if senators have a problem with the administration's handling of Benghazi they should "go after me" rather than try to "besmirch her reputation."

Graham also criticized U.S. intelligence agencies that wrote the talking points on which Rice based her public comments.

"I'm very disappointed in our intelligence community. I think they failed in many ways. But with a little bit of inquiry and curiosity, I think it would be pretty clear that to explain this episode as related to a video that created a mob that turned into a riot was far afield," he said. "And at the end of the day, we're going to get to the bottom of this."

Rice's controversial Benghazi statements were based on a set of unclassified talking points prepared by U.S. intelligence agencies for members of Congress.

The initial draft written by the CIA referred to "attacks" carried out by "extremists with ties to al Qaeda." However by the time Rice received them, "attacks" had changed to "demonstrations" and "with ties to al Qaeda" had been deleted, multiple U.S. sources have said.

The White House has denied making those edits and members of Congress are trying to determine where the changes were made.

The senators who met with Rice remained unconvinced by her responses and said her visit left them with greater concerns than before the meeting.

"I wouldn't vote for anybody being nominated out of the Benghazi debacle until I had answers about what happened that I don't have today," Graham said.

Asked whether he would block such a nomination, Graham said: "Oh, absolutely. I would place a hold on anybody that wanted to be promoted for any job that had a role in the Benghazi situation."

White House spokesman Jay Carney said the questions about Rice's appearance on the talk shows and the talking points had been answered. "The focus on - some might say obsession on - comments made on Sunday shows seems to me and to many to be misplaced," he said.

When you visit a review on Google Play for an app, book, song or movie, you won't have to worry as much about whether someone is sheepishly hiding behind a computer firing off nasty words, as Google has now flipped on the switch to display your Google+ name and profile picture with all reviews you add to the store. I'm not so sure this is the best thing in the world, 100%, as you don't have an option to turn it off. I like the idea of inviting people to share their thoughts, and mixing in real names and pictures if chosen by the user. That way, people know what reviews they might want to trust over other ones that are anonymous and thus, perhaps shady. The company seems to be choosing quality over quantity, though.

Being a step parent is not the easiest thing to do. Naturally, a child loves their parents, but when there is a divorce or the biological parents split things get complicated. When either parent finds a new partner they become the step parent. When a child has a new step parent they do not have that natural love like they do for their biological parents. The step parent has to earn that love and trust.
When you are a step parent you have many things that you have to take into consideration. How does the child feel? Is the child afraid of the new situation? How can you gain that love and trust? Is the child happy? These are just a few of the first questions you need to ask yourself when first becoming a step parent. You need to assess the situation and jump into gaining the love and trust of the child or children. But do not just into making them call you Mom or Dad, let them ask come to terms that you are in their life.
The step parent needs to be able to communicate with the child in a manner that lets the child know that they are there for them but not trying to take their biological mother or fathers place. This can be a hard thing to do but it is the most important. If the step parent comes through as trying to take over the parents role the child will become withdrawn and maybe even resentful of the step parent. Communication is the key to having a good relationship with the step child. One study shows that avoidance of and quality of conversation has a very big impact on relationships between stepparents and stepchildren. Most of all be very patient with a child.

We now know which two astronauts will have to pack their bags very, very well in the next three years: NASA's Scott Kelly and Roscosmos' Mikhail Kornienko have been picked for the year-long stay aboard the International Space Station in 2015. Both voyagers are old hands at space travel, having each spent a total of six months in orbit and at least some time on the ISS crew. There's no great shock in the choices when the mission will track the long-term effects of near-zero gravity on the human body -- after all, most of us would want a crew comfortable in its spacesuit boots for such an ambitious (though not record-setting) trip. Kelly and Kornienko will start a two-year training program shortly into 2013 that should have them in shape by the expedition's launch... and hopefully remind them to bring a good toothbrush.

Chemical 'switches' for neurodegenerative diseases discovered

ScienceDaily (Nov. 27, 2012) ? By using a model, researchers at the University of Montreal have identified and "switched off" a chemical chain that causes neurodegenerative diseases such as Huntington's disease, amyotrophic lateral sclerosis and dementia. The findings could one day be of particular therapeutic benefit to Huntington's disease patients. "We've identified a new way to protect neurons that express mutant huntingtin proteins," explained Dr. Alex Parker of the University of Montreal's Department of Pathology and Cell Biology and its affiliated CRCHUM Research Centre.

A cardinal feature of Huntington's disease -- a fatal genetic disease that typically affects patients in midlife and causes progressive death of specific areas of the brain -- is the aggregation of mutant huntingtin protein in cells. "Our model revealed that increasing another cell chemical called progranulin reduced the death of neurons by combating the accumulation of the mutant proteins. Furthermore, this approach may protect against neurodegenerative diseases other than Huntington's disease."

There is no cure for Huntingdon's disease and current strategies show only modest benefits, and a component of the protein aggregates involved are also present in other degenerative diseases. "My team and I wondered if the proteins in question, TDP-43 and FUS, were just innocent bystanders or if they affected the toxicity caused by mutant huntingtin," Dr. Parker said. To answer this question, Dr. Parker and University of Montreal doctoral student Arnaud Tauffenberger turned to a simple genetic model based on the expression of mutant huntingtin in the nervous system of the transparent roundworm C. elegans. A large number of human disease genes are conserved in worms, and C. elegans in particular enables researchers to rapidly conduct genetic analyses that would not be possible in mammals.

Dr. Parker's team found that deleting the TDP-43 and FUS genes, which produce the proteins of the same name, reduced neurodegeneration caused by mutant huntingtin. They then confirmed their findings in the cell of a mammal cell, again by using models. The next step was then to determining how neuroprotection works. TDP-43 targets a chemical called progranulin, a protein linked to dementia. "We demonstrated that removing progranulin from either worms or cells enhanced huntingtin toxicity, but increasing progranulin reduced cell death in mammalian neurons. This points towards progranulin as a potent neuroprotective agent against mutant huntingtin neurodegeneration," Dr. Parker said. The researchers will need to do further testing this in more complex biological models to determine if the same chemical switches work in all mammals. If they do, then progranulin treatment may slow disease onset or progression in Huntington's disease patients.
